Outline of Annual Research Achievements

細胞内外の多様な生命活動を支えるために、タンパク質は20種類もしくはそれ以上のアミノ酸で構成されるポリペプチド鎖からなる膨大な一次配列空間から適切な配列を選択し、多様な生体システムを形成している。こうした多様性を支えるためには、リボソームを中心としたタンパク質合成システムが多様な配列に対応できるような汎用性を備えていると考えられてきた。しかしながら、近年になって、こうしたタンパク質合成装置の高い汎用性に数多くの例外が存在すること、さらに、その例外が様々な生命活動において役割を担っていることが明らかにされており、核酸が持つ遺伝暗号がタンパク質のアミノ酸配列を規定しているという単純明快に見えたセントラルドグマで記述されるプロセスが、より複雑な階層構造を持ったシステムであることが示唆されている。本研究では、次世代シーケンサおよび再構成型無細胞タンパク質合成システムであるPURE systemを利用して、リボソームがmRNAの多様性に対して保持する一般性または特異性を包括的に解析し、セントラルドグマで記述されるプロセスにおける複雑な階層構造を定量的に記述することを目指した。これまでに、部位特異的にランダム化させた鋳型を用いた進化分子工学的アプローチによる選択系を用いて配列とタンパク質合成効率の相関の測定や、質量分析法を利用した1000種類強のペプチドの合成量を指標にした配列とペプチド合成量の相関を測定し、鋳型核酸配列とタンパク質合成効率の関係性を明らかにするいくつかのアプローチの構築に成功しており、データ解析によってタンパク質合成活性に高い影響を与える特徴的な配列の抽出を行うことが可能になった。
