Outline of Annual Research Achievements

本研究では、空間認知障害を引き起こすアルツハイマー病の症状が神経回路レベルでどのように進行していくか明らかにするために、空間の認識・記憶の障害に関係する海馬CA1領域に緑色蛍光カルシウムプローブG-CaMP7を発現するアルツハイマー病モデルマウス(AppNL-G-F/NL-G-F)を用いて、in vivo二光子カルシウムイメージングにより、同一個体の約650個からなる同一神経細胞集団の活動を数ヶ月に亘り観察し、神経回路レベルでのアルツハイマー病病態の進行に対して、いくつかの興味深い所見を見出した。(1)アルツハイマー病モデルマウスでは、海馬上昇層でG-CaMP7と共発現している赤色蛍光蛋白質DsRed2の蛍光シグナルとして観察されるアミロイド斑様凝集体が、3か月齢から発生し、加齢と共に数、大きさともに増大し、さらに、毒性の高いアミロイド付近に存在すること(2)神経活動においては、過活動を示す細胞が増加すること(3)場所細胞においては、安定性が4か月齢減少することと(4)数が7か月齢から減少すること(5)これらの異常な神経活動はアミロイド斑様凝集物の周囲で起こりやすいことがわかった。したがって、アルツハイマー病の海馬CA1領域の機能回路は、アミロイド蓄積が増加した後に場所細胞を含む神経機能回路が破綻していくと考えらえる。また本研究の実験系は、数か月に亘るアルツハイマー病症状の進行と細胞活動を同時観察できるため、新規アルツハイマー病薬の有効な評価に応用されていくことが期待される。
