Outline of Annual Research Achievements

筋萎縮性側索硬化症(ALS)や前頭側頭葉型認知症(FTD)においては、TDP-43やFUSなどのRNA結合タンパク質の凝集・蓄積を認める一方で、C9orf72遺伝子の異常伸長GGGGCCリピートRNAなどの凝集・蓄積も認めるが、両者を結ぶクロストークの実態は全く未解明である。
本研究ではRNA結合タンパク質によるリボスターシス維持機構を解明することを目的として研究を行い、以下の成果を得た。
異常伸長GGGGCCリピートRNAを発現するC9-ALS/FTDショウジョウバエモデルを樹立し、RNA fociの形成、リピート関連非ATG依存性(RAN)翻訳によるジペプチドリピート(DPR)の産生を認め、複眼変性、寿命短縮、運動障害などの表現型を呈することを明らかにした。正常鎖長のGGGGCCリピートRNAの発現では、特に表現型を認めなかった。続いてGGGGCCリピートRNAに結合するRBPのうち、RBP1の発現によりRAN翻訳が阻害され、表現型が抑制されることを見出した。以上の結果から、C9-ALS/FTD病態におけるRAN翻訳の重要性が示され、私たちのC9-ALS/FTDモデルショウジョウバエは、有用な動物モデルであると結論した。
