Outline of Annual Research Achievements |
微小管結合タンパク質タウは、アルツハイマー病を始め一群の神経変性疾患脳で様々な修飾を受け構造変化を起こして蓄積しており、それが神経細胞死を引き起こすと考えられている。しかしタウのどのような変化がその蓄積と神経細胞死につながるのかは明らかではない。 本研究では、そのタウの凝集と毒性の関係をin vivoで明らかにするため、凝集性が変異することが知られる変異を導入したタウを発現するトランスジェニックショウジョウバエを7種類作成し、それらによる細胞死の程度を比較した。すると、特にCys291とCys322 のAla置換変異(C291/322A)により、タウの神経細胞死が抑えられることを見出した。またC291/322A変異の導入により、タウのタンパク質量が減少しており、分解が早まることが示唆された。マウス初代培養神経細胞でも、C291/322Aタウは、野生型タウより分解されやすいことがわかった。培養神経細胞内で、タウはCys291とCys322を介してジスルフィド結合を形成していたので、ジスルフィド結合がタウの安定性に関わると考えられた。 Cys291/Cys322 はダイマー形成の促進によりタウ毒性に関わることが報告されている。しかし、本研究でみられたC291/322Aによる神経細胞死の抑制は、ダイマー形成よりむしろタウモノマーの安定性を減少させるという、新規な経路であることが示唆された。これらより、神経細胞内でタウのシステインを介したジスルフィド結合が増加することで、タウの量が増加し、細胞死が引き起こされることが示唆された。タウのジスルフィド結合は酸化ストレスによって増加するため、本研究はタウのジスルフィド結合が酸化ストレスと神経変性疾患発症をつなぐメカニズムである可能性を示した。
