Outline of Annual Research Achievements

ポリグルタミン病や認知症などの神経変性疾患では、分子シャペロンーユビキチンープロテアソーム系(UPS)やオートファジーの機能を凌駕して変異蛋白質が凝集して蓄積し、この過程に神経毒性が存在し神経変性が惹起される。有効な治療法は、病因蛋白質の異常な凝集を抑止したり、蓄積する量を減らすことで達成される。本研究では、オートファジーの制御メカニズム及びオートファゴソーム形成の分子機構の解明を目指して、オートファゴソームとリソソームの融合におけるdynactin1とTRAPPC9の役割を検討した。Two-hybrid法、免疫沈降法により、dynactin1とTRAPPC9はインターアクションした。運動ニューロン細胞NSC34細胞において、Western blot、RFP-GFP-LC3 レポーターアッセイ、電顕及びLC3の免疫電顕などの手法を用いて、dynactin1とTRAPPC9によるオートファゴソームとリソソームの融合に関して検討し、神経変性との関連も検討した。dynactin1とTRAPPC9をノックダウンすると、オートファゴソームとリソソームの融合が障害された。TRAPPC9はdynactin1、LC3-llやリソソームのマーカーであるLamp1とインターアクションした。また、dynactin1のノックダウンで、変異SOD1蛋白質の発現量が増加した。以上の結果から、dynactin1はTRAPPC9を介してオートファゴソームとリソソームの融合に関わり、神経変性の原因となる病因蛋白質の分解に関与することが判明した。Dynactin1及びTRAPPC9は、それぞれ分子量が150 KD程度の大きなタンパク質であり、様々な機能ドメインから構成され、TRAPPC9は他の種類のサブユニットと複雑なコンプレックスを構成している。今後は、これらの機能ドメインを同定することが課題である。
