Outline of Annual Research Achievements

小胞体カルシウム(Ca2+)シグナリングは、脳神経及びグリア細胞内のシグナル伝達を担う。認知症の素過程と考えられる「脳タンパク質老化」が、この小胞体Ca2+シグナリングと如何に相互作用するか研究を行っている。これまで小胞体Ca2+チャネルがオートファジーと小胞体ストレスを調節することを発見し(BBA 2018)、小胞体Ca2+チャンネルの動作原理を解明した(Messenger 2018; BBA 2018)。アルツハイマー型認知症の発症に関与するTauタンパク質のアイソフォームを培養細胞に過剰発現し小胞体Ca2+放出を測定する実験を行った結果、タウタンパク質(Tau)が小胞体Ca2+放出に作用することを見いだした。他の神経変性疾患の発症に関与するαシヌクレイン(Syn)やポリグルタミン(polyQ)も同様に小胞体Ca2+放出に作用するので、Tau/αSyn/polyQに共通の調節メカニズムが示唆された。更に、小胞体Ca2+放出がこれらのタンパク質に及ぼす影響を培養細胞で調べた結果、興味深いことにタンパク質老化を引き起こす構造変化を検出した。本研究により初めて得られた小胞体Ca2+シグナリングとタンパク質老化との相互作用は、認知症の素過程を知る重要な鍵となると考えられる。今後はこの分子メカニズムを解明し、認知症の治療や予防に役立てたい。
