Outline of Annual Research Achievements

脳の様々な領域はカラム構造と呼ばれる機能単位から成るが、その形成機構はほとんど分かっていない。ほ乳類においてカラムは数万の神経細胞から成り、その全貌を明らかにすることは困難であるが、ショウジョウバエ脳視覚中枢において見られるカラム構造は100程度の神経細胞から成る上、ハエの分子遺伝学的手法を用いて迅速に解析を進めることができる。本研究ではハエ視覚中枢を用いてカラム形成の分子機構を解明した。以下の1~2については成果を国際的な学術誌に掲載し、3~5については投稿した論文がリバイズ中となっている。
1. R7, R8, Mi1の3種類の神経細胞がカラム形成において中心的な役割を果たすことを明らかにした。2. 細胞接着分子Nカドヘリンによる細胞接着力の違いによってR7, R8, Mi1のカラム内での位置関係が制御されることを明らかにした。3. カラム形成過程において同じ神経幹細胞に由来する神経細胞どうしが反発し、異なるカラムに投射する「細胞系譜依存的反発」現象を見いだした。4. ダウン症の原因遺伝子とされるDscam1が神経幹細胞において一過的に発現し、同じ細胞系譜間の反発を誘導することで、細胞系譜依存的反発およびカラムの形態形成を制御することを明らかにした。5. ハエ視覚中枢の腹側および背側特異的に発現するWnt4およびWnt10が濃度勾配を形成し、その制御下において、受容体として働くFz1/Fz2およびPCPシグナル因子が働く事で、3次元構造であるカラムの極性が形成することを明らかにした。
