Outline of Annual Research Achievements

発達期に特定の周波数の音を継続的にマウスに聴かせると、大脳皮質一次聴覚野(AI)にその周波数の音に反応する領域が周波数軸方向に拡大するが、この可塑的変化に聴覚皮質がどのように関与するか、一次聴覚野以外の領野にも可塑性が見られるかは不明である。本研究では、これらの問題を解明することを目的とした。まず、聴覚皮質の発達の様子を解明した。光イメージング法を用いて、生後14日(P14)からP60まで、AIと前聴覚野(AAF)の発達の様子を可視化し、発達に従って、1)両者の周波数軸がなす角度が小さくなることと、2)両者の周波数軸間の距離が長くなることを明らかにした。これらのことから、AIとAAFは生後発達に従って、両方とも大きくなることが分かる。一方、上記のように機能的に同定したAIとAAFの境界をマークし、組織切片において検査したところ、その境界は海馬の先端に対応することを見出した。この結果は、脳スライス標本において、AIとAAFを同定するために重要である。可塑性を誘導するために、P12からP15まで中間周波数の純音に暴露すると、AIとAAFの境界が移動することを見出し、AIのみならず、AAFにも経験依存的な可塑性が存在することが示唆された。可塑性に伴う聴覚皮質内の詳細なメカニズムを知るためには、特定の周波数に応答する皮質細胞を標識する必要があるが、活動依存的分子マーカーをエレクトロポレーション法を用いて、第2/3層の皮質細胞に導入し、標識することに成功している。この方法とマルチパッチ法を組み合わせて、脳スライス標本において、可塑性に伴う聴覚皮質内の詳細なメカニズムに関する解析を進めている(Luo et al., 2017)。本年度に聴覚野に関する論文を発表する(Nishimura et a., 2018)と同時に、関連論文も発表した(Wu et al., 2018)。
