Outline of Annual Research Achievements

末梢神経の損傷は、その投射路の上位中枢で神経回路の改編を引き起し、これは脳内マップの改変や幻肢痛にも深く関わることが知られている。我々は、視床VPM核へのヒゲ由来の上行性の内側毛帯線維シナプスを他と識別化することで、ヒゲ感覚神経を損傷したマウスのVPMにおいてヒゲ由来の内側毛帯線維シナプスが弱化し(シナプス刈り込み)、ヒゲ以外の体幹や下顎由来の内側毛帯線維シナプスが、ヒゲ領域ニューロンに下顎などからの異所性シナプスをつくることを見いだした。本研究では、視床のシナプス刈り込み・異所性シナプス形成といったヒゲ感覚神経損傷によって引き起こされる二種類のシナプス改編機構に対し、ミクログリアが特異的に作用するかを検討し、それにより体部位マップ変化や異所痛行動に与える影響を調べた。その結果、ヒゲ感覚神経損傷後5日目で三叉神経核ヒゲ領域のミクログリアは集積して数が増え、活性化していた。一方、視床ではミクログリアの数も形も変化なく、CD68で活性化を見ても、三叉神経核のミクログリアは活性化されており、視床は活性化していなかった。加えて、ミクログリアを欠損させたマウスを用いると、神経損傷後の視床シナプスの改編は阻害された。三叉神経核のみミクログリアを除去しても同様であった。以上のことから、起始核でのミクログリアがシナプス前に働いて神経損傷に伴う軸索の改編を起こしていることが示唆された。
行動レベルでは、ヒゲ神経を切断すると顎の触刺激に対するアロディニアが生じることをすでに報告しているが、シナプス改編現象と一致して、ミクログリアを欠損させるとアソレニアが阻止されることが分かった。以上のことから末梢神経損傷による起始核のミクログリアの集積・活性化が視床でのシナプス改編とアロディニアを誘発していることが示唆された。
