Outline of Annual Research Achievements

網膜は6種類に大別される神経細胞とミューラーグリアで構成され、発生の時間軸に従って、各細胞系列が逐次的に共通のプロジェニター細胞から分化を遂げる。マウスと人の発生の時間軸が、網膜においても異なることを利用して、この時間軸の制御にヒストンH3K27のメチル化がどのように拘るのかを検討することを目的とした。human, mouse iPSの混合網膜誘導培養の系の樹立を試みていたが、それぞれ異なる条件で培養する細胞の混合培養は想定より技術的な困難が伴うことが明らかになったためこの系の開発は断念し、より直接的にヒストンメチル化の網膜発生時間軸の制御に果たす役割を解析することとした。このため、これまで網膜最終分化のフェノタイプ解析が主体であったH3K27me3メチル化酵素網膜特異的ノックアウトマウスについて、詳細な分化過程のフェノチアプの解析に加え、すでに整備したH3K27me3のChIP-seqデータの細胞系列特異的転写因子、サイトカイン類などの遺伝子座についての修飾状態を把握し、細胞系列、発生段階に加えて、遺伝子機能での検討を行いH3K27me3の網膜発生の時間軸制御について、新たな仮説を得ることができた。研究班内で関連遺伝子の新たなマウスモデルの提供を受け、この仮説の検証を行い、ヒストンH3K27me3の脱メチル化が発生にクリティカルな転写因子の遺伝子座の脱メチル化を行うタイミングを制御し、網膜発生時間軸を制御していることが示唆された。
