Outline of Annual Research Achievements

霊長類などの高度な機能を発揮する脳構築の特徴である脳回を持つ大脳(複雑脳)の形成過程で、脳室帯と呼ばれる普遍的な幹細胞帯と、発生途上で新たに形成され神経幹細胞層(外幹細胞帯)で、神経幹細胞のidentityの時間的進行が同じように進むのかどうかという課題に取り組んだ。モデル系として単純ながら複雑脳の特徴を持つフェレットを用いた。幹細胞帯が脳室帯と外脳室帯に別れた後も、基本的には、ニューロンを作り出す幹細胞identityの時間変化はほぼ同時に進行していることが判明した。しかし、神経層にまでニューロンが到達するのに要する時間が大きく違うため、同じbirth dateのニューロンでも、誕生した場所が脳室帯か外脳室帯(outer subventricular zone)かによって、異なる神経層に到達してることが判明した。一方で、フェレットは誕生後も神経発生が生後10日ぐらいまで引き続くが、体性感覚野では、脳室帯と外脳室帯ではこの時点から大きくことなる事態が進行することも判明した。外脳室帯はそのまま神経産生を続け、グリア前駆体も生じてくるのに対して、脳室帯では、細胞分裂が減衰し、上皮形態を維持したradial glia (RG)とtruncated RG (tRG)が共存する。tRGと考えられる細胞は脳室帯の脳室面から離れた領域で整列が進行し、数層のキューブ状(核も細胞体も)の細胞の列が形成される。これらの細胞は違いに接着分子で密に接着し脳室面に太いendfootを伸ばしている。これらの多くの細胞がCryab陽性ついでFoxj1陽性となり、その後、核が脳室面に移動し、主にependimal cellになることを見出した。このような細胞体の顕著な整列を含む分化プロセスはマウスでは見られないものであり、脳回を持つ哺乳類の脳に特徴的な現象の可能性がある。
