Outline of Annual Research Achievements

我々は、制御性T細胞(Treg)による自己免疫寛容の確立・維持メカニズムを明らかにするために、ヒト自己免疫疾患IPEX症候群患者に見つかっているFoxp3変異を導入したマウスモデルを作製・解析してきた。その結果、Foxp3<A384T>変異により皮膚、肺などのバリア組織選択的に2型および3型の炎症が惹起されることを見出した。Foxp3<A384T>変異マウスが何故組織選択的な自己免疫疾患を発症するのかを明らかにするために、この変異によってTregのTCRレパトアに組織選択的欠損(“穴”)が生じ、このためにこれらの組織においてのみ免疫寛容が破綻して炎症が惹起されるという仮説を立てた。この仮説を検証するために、TregおよびTconvのT細胞受容体(TCR)レパトア解析を行うこととし、レパトア解析のために既に遺伝子再構成を受けたTCRbeta遺伝子を持ち一方のTCRalphaアレルのみ発現できる1D2b x TCRCa+/- x Foxp3<A384T>変異マウスを交配により樹立した。まず、TCRβ鎖を固定した場合としない場合とでFoxp3<A384T>マウスの表現型に違いが見られるか検討したところ、どちらの場合でも皮膚と肺に炎症が見られ、肝臓では見られなかった。興味深いことに、TCRβ鎖を固定しない場合の皮膚炎は全体的に軽度で個体差が大きいのに対し、TCRβ鎖を固定することにより全ての個体で皮膚炎のみが特異的に重症化することを見いだした。一方、Foxp3<WT>背景ではTCRβ鎖を固定しない場合はもちろん、固定しても皮膚およびその他の臓器に炎症は発症しなかった。以上の知見は、TCRβ鎖を固定することによるTCRレパトアの偏り(多様性の減少)がFoxp3<A384T>変異と相互作用することにより、発症する自己免疫疾患の組織特異性が大きな影響を受けることが明らかになった。
