Outline of Annual Research Achievements

腫瘍組織に浸潤するB細胞とT細胞の細胞数は患者の予後と正の相関を示すことが報告されている。腫瘍浸潤B細胞(TIB)は、抗原受容体のクラススイッチ・親和性成熟を経ていることから何らかのがん抗原に反応し活性化しており、また、がん抗原をT細胞へ提示していると思われる。そこで、がん患者の腫瘍組織に浸潤するTIBを単離し、CD40リガンドとBAFを発現させたフィーダー細胞上で培養して、さらにBach2を遺伝子導入することによって長期に培養可能な方法を確立した。また、Bach2の活性を抑制するhemeを培養に添加することにより、このTIB細胞の形質細胞への分化を誘導し抗体産生を促進する方法も見出した。こうして培養上清中に得られた抗体を用いて、TIBの元のがん組織切片を免疫染色したところ、腫瘍細胞が染色される例が見出された。これらTIB細胞をクローン化し、がん抗原に特異的な抗体を産生するTIBクローンを選択している。
また、このTIBクローンをマウス個体に移入し、個体内で抗体産生細胞に分化させることにより、がんを標的としたB細胞療法を計画している。この目的のために、タモキシフェンで制御可能なBach2-ERT2融合遺伝子を作製し、これをTIB細胞に導入した。このTIB細胞はタモキシフェンを添加しない培養では形質細胞に分化した。したがって、この細胞を個体に移入すれば体内で抗体を産生すると思われる。
