Outline of Annual Research Achievements

本研究の目的は、生殖器官の形成過程における転写因子とエピジェネティック因子複合体の作用機構の解析を通して、時空間特異性の制御機構を理解することである。これまでに花発生過程において、幹細胞の増殖停止に機能する転写因子の複合体に着目し、それらのターゲット遺伝子の選択性を解析してきた。転写因子であるSUPERMANは花発生初期において、花幹細胞群を取り囲むように発現している。SUPタンパク質は、ポリコム因子であるFIEと直接結合して、オーキシン合成酵素YUUCCA (YUC)遺伝子の発現抑制をおこなっている事、実際にオーキシンの生合成を介して花幹細胞の増殖を抑制していることを示した (EMBO Journal, 2018)。また、CRC転写因子はSUPから2日ほど遅れて、幹細胞を取り囲むように発現する。CRCもまたYUCを直接制御しており、CRCの上流因子であるAGとともにYUC遺伝子座に直接結合する。すなわち、AGはまずクロマチンリモデリング因子の導入により、クロマチン構造を開き、その後にCRCが直接結合することで、YUCを活性化している。そのクロマチンを介したオーキシン合成酵素の制御機構の論文をNature Communications (2018)にて発表した。さらに、細胞自立的に花幹細胞の増殖抑制にかかわる経路の解析を行い、KNUがクロマチンリモデリング因子の結合阻害と抑制的ヒストン修飾酵素の導入という多段階のエピジェネティック制御を介した遺伝子ネットワーク機構を解明して論文報告を行った(Plant Cell 2019 in press)。以上、花幹細胞の増殖抑制の時空間性の制御として、転写因子とヒストン修飾酵素との精緻なクロストークによるエピジェネティク制御機構を解明することができた。
