Outline of Annual Research Achievements

同質受精卵および異質倍数性受精卵の発生機構を明らかにすることを目的に、主に下記3項目の研究を遂行した。
1.イネ受精卵発生における父性鍵因子の同定:受精後に父性アリル依存的に発現が誘導される遺伝子群を同定し、これらのうちAP2型転写因子であるOsASGR-BBML1をイネ卵細胞に異所的に発現させたところ、卵細胞が受精非依存的に核分裂・細胞分裂し、細胞塊にまで増殖した。さらに、受精卵中のBBML1の機能抑制を行ったところ、受精卵は第一分裂することなくその発生を停止した。これらの結果から、OsASGR-BBML1が受精卵発生の鍵因子である可能性が強く示唆された。
2.交雑受精卵の発生機構:コムギin vitro (IVF) 受精系を新たに確立し、イネIVF系と組み合わせることにより交雑受精卵(異質倍数性受精卵)の発生プロファイルを正確に観察・解析することを可能にした。イネ卵細胞とコムギ精細胞を融合させて作出した受精卵は球状胚まで発生を進めた一方で、雌雄を逆にした受精卵は核合一後に分裂をせずに発生を停止した。また、複数のイネ卵細胞と1個のコムギ精細胞を融合させた異質倍数性受精卵では、球状様胚からさらに発生が進んだ増殖細胞塊にまで発生を進めること、さらには、イネ-コムギ複合受精卵は植物体へと再分化した。これらの結果は、受精卵中における異種雌雄ゲノムの存在比とその交雑受精卵発生の関係性を明確にし、さらに、異種の雌雄配偶子の組み合わせの至適化により交雑不全を克服することができる可能性を強く示している。
3.単離卵細胞の受精非依存的な分裂・発生を誘導する化合物のスクリーニング:ヒストン脱アセチル化酵素阻害剤が卵細胞の自律的核・細胞分裂を引き起こすことを見出した。さらに、当該阻害剤の処理時間を調整することで、分裂率が向上するとともに、卵細胞がカルス塊にまで増殖することを明らかにした。
