Outline of Annual Research Achievements

膜蛋白質として、イオン透過型グルタミン酸受容体の一つであるAMPA受容体について、電子顕微鏡解析用にGSG1LおよびStargazinとの融合蛋白質の作製を行った。界面活性剤可溶化状態での精製後の脂質ナノディスクへの再構成、リポソームへの再構成後に電子顕微鏡による確認を行ったが、現在までのところ良好な結果は得られていない。現段階では、電子顕微鏡での試料の性状確認で良好な結果が得られず、AFMでの解析についてはと計画段階で停滞している。
方法論開発として先行して行っていた電依存性カリウムチャネルKv1.2を脂質小胞に再構成した系では、分解能を向上させるべくより高性能な電子顕微鏡の利用を検討し、米国Yale大学の電子顕微鏡の利用を開始したが、これまでに分解能の向上は得られていない。それまでに使用していたアモルファスカーボン膜によるノイズが問題となり分解能向上が見られないと考え、炭素数層からなるグラフェンを支持膜として用いる系に移行した。理研播磨でのグラフェン作製を試み、電子顕微鏡グリッドへの転写、プラズマ装置による洗浄、生体分子吸着を向上する水素化処理を検討した。転写の効率は高くなったが、まだ、プラズマによる洗浄が十分でない状態である。並行して市販のグラフェン貼り付けグリッドを水素プラズマ処理をし、標準試料での2.5オングストローム構造を得られており、この系をAMPA受容体に適用する。
