Outline of Annual Research Achievements

P1ニューロン群は、我々が世界で初めてキイロショウジョウバエにおいて同定した雄の求愛開始意志決定ニューロン集団である。P1ニューロンは、片半球あたり20個の雄特異的介在ニューロンで形成されている(Kimura et al., 2008, Neuron 59, 759-769)。実際、本来は雄特異的はP1ニューロンを雌の脳内に人為的に発生させれば、その雌が雄の求愛行動をするようになる。また、野生型成虫羽化後の集団生活経験による雄の求愛行動の抑制も、P1ニューロンの機能的可塑性に依存して生ずる。現在、複数の研究グループの努力によりP1ニューロンに求愛制御のほぼすべての情報が収斂して統合され、P1が主要な求愛決定中枢として働くことが確立しつつあるが、このようにP1がこの特権的機能を発揮できる理由は不明である。野生型の雄を成虫羽化後、数日間隔離して育てると、複数の雄の集団の中で育てた場合に比べて雌への求愛が著しく増強される。この背景にP1ニューロンの行動惹起閾値の低下(受けた刺激が弱くても行動出力をトリガする)があることは、optogeneticsとCa2+ imagingによって示唆されていた(Inagaki et al., 2014, Nat. Methods 11, 325-332)。我々はP1にin vivo patch clampを施し、膜電位固定下で記録される膜電流のうち遅延整流性外向きK+電流(ID)のみが、特異的に集団生活によって、一部のP1ニューロンで劇的に増加することを明らかにした。このことから、羽化直後からの数日を臨界期(臨界点)として社会経験を膜電流に刻印するP1ニューロンが存在し、その働きで求愛行動の経験依存的変容が生ずると推察された。
