Outline of Annual Research Achievements

生体を構成する細胞は多様な外部環境に適応し、細胞分化し、機能を果たす。この細胞分化誘導にHippo-YAPシグナル伝達経路やMKK7-JNK経路が関与することが報告され、国内外の注目を集めている。我々は、Hippo-YAP経路がメダカ初期胚の細胞張力を制御することを報告してきた。また、MKK7-JNK経路がマウス発生期の脳形成に必須の役割を果たすことを示してきた。本研究では、課題1:細胞外硬度差に応じて、Hippo-YAP経路の細胞分化誘導するメカニズムの解明と、課題2:MKK7-JNK経路の成体マウスの神経系における役割の解明を目指した。その結果、課題1は海外のグループに先行されてしまったため、当初予定を修正して、研究を継続している。一方、課題2は興味深い結果を得ることができ、論文報告した。神経細胞特異的にMKK7を欠損するマウスは、1)概日リズム異常の行動を示すこと、2)加齢依存的に行動異常となること、3)この原因として加齢依存的な筋萎縮が生じること、が明らかとなった、すなわち、MKK7-JNK経路は、発生期では脳の形成において必須であり、成体では神経機能維持に必須の役割を果たすことが示唆された。現在、神経細胞内でのMKK7-JNK経路の役割を分子レベルで明らかにする研究が進行中である。また、MKK7欠損マウスは、高齢者社会で問題となっているサルコペニアのモデルになる可能性があり、その検討を行っている。さらに、4)ゼブラフィッシュを用いて、細胞時計の解析を行った。光誘導性の時計遺伝子が細胞時計の同調とゼブラフィッシュの行動量の制御に関与することを明らかにした。2つのシグナル伝達系の研究から、大きく研究が展開する結果となった。
