Outline of Annual Research Achievements

本研究課題では、二量体型生物活性中分子アルカロイドの効率的合成実現のため、合成終盤でのカップリングに耐えうる官能基共存性に優れた画期的な方法論を確立し、誘導体合成を視野に入れた全合成を達成することを目的としている。最終年度の本年度は、これまで、二量体型インドールアルカロイド、ハプロファイチンの合成研究の過程で見出したAgNTf2を用いたFriedel-Crafts 型のカップリング反応を応用し、ピロロインドールのベンジル位とインドールN１位が結合した二量体構造の構築に成功した。通常、インドールの求電子置換型の反応は３位あるいは２位で起こることが知られている。今回、オルトアルキニルアニリンとブロモピロロインドールとのAgNTf2存在下の反応を行ったところ、ピロロインドールのアミノ化にカップリングと続く5-endo-dig形式の環化反応が段階的に進行し、目的の二量体を得ることができた。本反応を応用し、二量体型中分子アルカロイドである、(+)-pestalazine Bの全合成に成功した。さらに、フタロシアニン触媒による酸素酸化を利用した、Late Stage Oxidative Coupling戦略に基づく二量体型インドールアルカロイドメリナシディンIVの全合成の達成に向けた検討として、合成終盤でカップリング反応を検討した。また、スピロ環を介して２つのアスピドスペルマ型インドールアルカロイドユニットが二量体型構造をとっているボブツシンのモノマーユニットである(-)-deoxyapodineの合成経路の最適化を行い、これまでで最も短工程での全合成を達成した。また、生薬トコンに含まれる二量体型イソキノリンアルカロイドであるemetineは、かつて催吐剤として臨床利用されていたが、今回、有効な薬剤の無い肺高血圧症の新規薬剤として有効であることを見出した。
