Outline of Annual Research Achievements

近年、ペプチドや核酸を基盤とする中分子が次世代医薬品の候補として熱い視線を集めている。研究代表者は、細胞内輸送小胞(エンドソーム)に保持された抗体を効果的に細胞質に放出可能とする「エンドソーム不安定化ペプチド(L17E)」を創出した。本研究では、L17Eの細胞内移行機序を理解するとともに、種々のL17Eの誘導体の合成を通じて、生理活性タンパク質や生物機能中分子の細胞内送達活性向上を目指した。その結果、(1)L17Eの膜透過促進作用は、エネルギー依存的であり、エンドサイトーシスの極めて早い段階で発揮されることを見出した。この機序は、従来から知られてきた直接膜透過や成熟したエンドソームからの脱出とは異なる、新しい概念による細胞内送達機序であり、生物機能中分子の細胞内送達にも有用であると考えられた。さらに、(2)溶血ペプチドの膜傷害性の調節により、新しい細胞透過ペプチド(弱毒化溶血ペプチドn22E2K2、ChryE2)を開発した。これらの細胞透過ペプチドと、生物機能中分子としてのモデルペプチドとの架橋体を調製することにより、モデルペプチドの効果的な細胞内送達と細胞内活性の発現が確認された。さらに、(3)新規マクロピノサイトーシス誘導ペプチドSN21と溶血ペプチドとのコンジュゲートを用いて、高効率の抗体を含むタンパク質や、siRNA、プラスミド等の核酸類の細胞内送達が可能であること示された。本年度の研究成果を4報の論文として公表するとともに、国内外の学会で成果の発表を行った。また、特許出願を行った。
