Outline of Annual Research Achievements

P糖タンパク質(P-gp/MDR1/ABCB1)とBCRP/ABCG2は、代表的なトランスポーターの2つである。多くの薬物は、この二つのトランスポーターによって、排出輸送を受ける。そこで、2019年度は、先ず、複数のトランスポーターによって作り出される輸送分子夾雑系を解明する手法の確立のためのモデルとして、この2トランスポーターに焦点を当て、in vivoの輸送分子夾雑系を解明できるかどうかを検証した。P-gpとBCRPについて、高発現させたin vitro細胞株とその親細胞株(発現させていない)を用いて、典型的な基質に対する輸送速度を計測し、高発現株と親細胞株の差からトランスポーターの輸送活性を求めた。液体クロマトグラフィ接続型質量分析装置を用いて、その細胞株における該当トランスポータータンパク質の存在量を定量した。輸送活性を存在量で割ることによって、1 molあたりの輸送活性を解明した。1 molあたりの輸送活性を、マウスの血液脳関門におけるトランスポーターのタンパク質存在量(mol)と統合することによって、in vivo血液脳関門での輸送活性を再構築した。実測値(マウスで計測)と比較した結果、良好にin vivoの血液脳関門の透過速度を再構築できていることが実証された。ヒトへの応用性を検証するため、ヒトに近いカニクイザルで同様の検証を行った結果、良好な結果が得られたため、ヒトの血液脳関門の輸送分子夾雑システムを解明するための方法が確立された。酸化ストレス等の病態時の血液脳関門の分子機構も解明し、トランスポーターの輸送機能を構成する因子を解明できた。独自に開発した網羅的定量プロテオミクス技術によって、正常および中枢疾患時における血液脳関門で発現する輸送担体の種類と量を一挙に把握でき、上述の再構築法と組み合わせることで、ヒト血液脳関門の輸送システムを定量的に理解できるようになった。
