Outline of Annual Research Achievements

我々の体の最小単位である細胞は、多数の分子が相互作用してある特定の機能を発揮する閉鎖空間であり、この複雑な分子の相互作用ネットワークを円滑に動作させる役割を直接的に担っているのが酵素分子をはじめとしたタンパク質である。本研究では、細胞を1細胞ごとに細胞と同程度の大きさを有するマイクロチャンバー内に隔離し、ナノメートルサイズの鋭い先端を有するナノワイヤを用いて細胞膜を破砕した後も細胞内の分子ネットワークの流れ、すなわち酵素分子の機能(酵素活性)が保たれるかどうかに焦点を当て、モレキュラークラウディング剤で再構成した疑似細胞内との比較検討も加えることで、細胞内の分子夾雑系が細胞機能に与える生物学的意義の本質的理解を目指した。
本年度は、まず蛍光タンパク質と酵素分子の共発現系の構築を行い、簡便に1細胞内の酵素分子数と酵素活性を同時に評価できる実験系を確立した。さらにマイクロチャンバー内にあらかじめモレキュラークラウディング剤であるPEGを封入することで、細胞膜破砕後も擬似的な細胞環境を再現することに成功した。これら疑似細胞内環境の構築は分子夾雑系定量評価のための基盤となるためのものであり、本研究においてはじめて実現したものである。今後は、実験データの蓄積を通じて分子夾雑系の本質的な理解をすすめるとともに、酵素分子の機能制御を可能とするモレキュラークラウディングをはじめとした疑似細胞内環境の合理的な設計指針を確立していく。
