Outline of Annual Research Achievements

ヒトの腸管には100兆個もの細菌が共生しており、この「腸内細菌叢」は未消化物を発酵分解し、多種多様な代謝物を産生することで、外付けの臓器とも呼ぶべき代謝系を構築している。腸内代謝バランスに影響を与える因子として食事に含まれる脂質が挙げられる。とくに脂肪酸の質の違いは炎症性腸疾患の発症・再発に関わるとされているものの、これらは経験則や疫学調査に基づくものであり、その分子メカニズムについては分かっていない。申請者はこれまで、腸内細菌が産生した脂質代謝物には、宿主側の免疫応答を制御する活性が認められることを見出している。本研究では、宿主と共生細菌の相互作用が織りなす「生物間代謝経路」から生じる脂質代謝物に着目し、免疫調節活性物質を探索した。マウス糞便中に含まれる脂溶性代謝物の免疫調節作用を明らかにし、その活性本体の単離を試みた。まず免疫調節活性画分を絞り込み、リピドミクスを実施した結果、複数の脂質代謝物が含まれていた。これら代謝物のうちの一つが制御性T細胞誘導活性を示すことが判明した。この代謝物は、T細胞単独培養系では作用を発揮せず、樹状細胞との共培養系でのみTreg誘導作用を発揮したことから、その作用は樹状細胞を介することが明らかとなった。無菌マウスでは便中の本代謝物はほぼ検出されないことから、腸内細菌が主要な産生源であることを確認した。さらに、本代謝物の便中濃度は大腸炎誘導時において顕著に低下することから、炎症時に減少する偏性嫌気性菌によって産生されていることが示唆された。
