Outline of Annual Research Achievements

染色体分配機能に必須なセントロメアでは、反復DNAからなる巨大領域にヒストンH3バリアントであるCENP-Aクロマチンが集合し、これを足場に100を超える因子群からなるキネトコアが形成される。キネトコアでは微小管との相互作用により染色体の動きを制御する。一方、セントロメア内部ではヘテロクロマチンが集合し姉妹染色分体の接着・分離を調節する。しかし、同一反復DNAの巨大領域にどのようにこれらの異なるクロマチンが集合し、機能統合体を保つのかは不明なままである。本研究では、セントロメアの階層的集合メカニズムの解明を目的とし、ヒト人工染色体(HAC)やChIP-seq法を用いた解析によりR1年度は以下の成果を得た。(1)染色体機能形成の階層性とヘテロクロマチンの解析:CENP-BはCENP-Aクロマチンの集合促進・安定化に関わるが、これとは拮抗するヘテロクロマチン化も促進する。これまでにCENP-B結合の有無により反復DNA上へリクルートされるヒストン修飾酵素やクロマチン関連因子をスクリーニングし、ヘテロクロマチンタンパク質HP1や、これとは逆にopen側修飾に関わるH3K36me2,3化酵素ASH1Lを同定した。CENP-Bはゲノム反復DNA上でASH1LとHP1の拮抗的な集合のバランス調節に関わっていることを明らかにした(Otake et al, 論文投稿中)(2)ゲノム反復DNA上のクロマチン構造の解析:白髭班と協力してヒトゲノムデータベースhg38を用いてゲノム反復DNA上のクロマチン構造について、ChIP Seq法により解析を進め、CENP-Aクロマチン、オープンクロマチン(H3K4me3、H3K36me3)、ヘテロクロマチン(H3K9me3、H3K27me3)等の分布をCENP-BのWT株とKO株で比較解析した(Kugou et al、 論文準備中)。
