Outline of Annual Research Achievements

(1) Rorippa aquaticaのゲノム解析: R. aquatica のゲノムをイルミナおよびPacBioプラットフォームでシークエンスし、また、Hi-C法により染色体スケールのスキャフォールドの作成に成功した。染色体間の配列比較により、R. aquaticaは異質倍数体で、染色体の一部が融合していることが示された。イヌガラシ属植物の2種の雑種からR. aquaticaが成立したと考えられる。
(2) 水没による葉形の制御機構の解析:R. aquaticaは、水没や温度の変化、光環境の変化で葉の形態が大きく変化する。水没時のRNA-seq解析により、水没時にはエチレン関係の遺伝子群の発現が変動していることが明らかになった。実際、エチレンを添加すると、葉の形態が水中葉に近くなることから、エチレンが水没応答に関わっているものと考えられた。
(3) 水没による気孔形成の抑制機構の解析:R. aquaticaの気孔形成は水没により抑制されるが、気孔形成に重要な転写因子であるSPCHの発現抑制が起こっていることを明らかにしていた。さまざまな条件における水没後の発現応答をRNA-seq解析で調べたところ、気孔形成の抑制時に関わる転写因子等を同定できた。SPCHの発現制御機構に着目して、これらの因子の役割を明らかにすることで、水没による気孔形成の抑制機構が明らかにできると考えている。水没時の光条件などを検討したところ、暗所では水没させても気孔形成は抑制されず、また、青色光のみを当てた場合も気孔形成は抑制されなかった。また、気中条件であっても赤色光を当てると気孔の形成が抑制された。以上の結果から、光質などの光条件が水没の感知に重要であることが示唆された。
なお、当初、学会等への参加のための旅費の執行を予定していたが、新型コロナウイルスのため中止になったため、未使用金が生じた。
