Outline of Annual Research Achievements

水分ストレス依存的に、根から地上部に移動するペプチドを受容する2つの受容体に着目し、それら遺伝子の組織特異的発現解析を行った。1つの受容体はプロモーターGFP植物を用いた解析から、葉の葉肉細胞および維管束組織の両方で発現していることを明らかにした。さらに、根の細胞に着目したシングルセル発現解析の手法を用いて、もう一つの受容体の発現組織の解析を行った。その結果、この受容体は維管束組織でのみ発現していることを明らかにした。ペプチドによるABA合成制御シグナルは、2つの受容体が破壊された2重変異体でのみ消失することを考えると、これら2つの受容体が発現している維管束組織が、ペプチド-受容体シグナルが成立する組織であることが明らかとなった。また、シロイヌナズナのLRR-RLK型受容体は約600遺伝子以上存在すること、さらに、これら多数の受容体は、主にヘテロ2量体を形成することが示唆されている。本研究成果は、ヘテロな受容体の組み合わせが、植物体内で実際に機能すること、さらに複数ある受容体群が様々なヘテロな組み合わせを形成することで、その受容シグナルを適切に認識し、下流へ伝達し分けていることを示唆する知見となった。また、受容体2重変異体を用いて網羅的な発現解析を行い、ペプチド-受容体群が制御する下流シグナル伝達経路の全容を明らかにする試みを行った。その結果、ペプチド-受容体はABA合成酵素の発現だけでなく、ABAが関わらない浸透圧応答に含まれる遺伝子群も制御していることを明らかにした。特に、浸透圧ストレス耐性に関わる転写因子群だけでなく、アミノ酸や糖の合成・代謝に関わる酵素群も多数、制御されていることを明らかにした。これらの結果から、ペプチド-受容体は、ストレス応答に関わる代謝産物の蓄積も制御して、水分ストレス耐性を個体全体で獲得させる重要なシグナル因子であることを明らかにした。
