Outline of Annual Research Achievements

カブトムシの角やクジャクの羽といった性選択形質は,ひときわ目を引く巨大で美麗な“誇張された表現型”であり,こうした武器や装飾はすさまじい種内・種間の多様性を持ち,進化的・発生的に“変わりやすい形質”であることが知られている.一方,節足動物の交尾器や哺乳類の脳など,他の体モジュールは生育時の環境によらず,ほぼ一定のサイズに発生し,発生プロセスも環境や遺伝的摂動に対して頑健な作りになっていると考えられている.私は,武器をもつ昆虫をモデルとして,様々な体モジュールが,全体として機能的なパフォーマンスを持つよう統合的に発生する仕組みに興味を持っている.本研究では,オオツノコクヌストモドキという武器甲虫を主なモデルとして,武器をもたらす遺伝子,そして武器サイズに大きな“ゆらぎ”を許容する分子基盤の解明を目指す.
武器の誇張化メカニズムおよび,武器と他の体モジュールに発生的リンクをもたらすメカニズム解明にむけて,生態発生学的解析を進めた.具体的には,ターゲット遺伝子に的を絞った解析(candidate gene approach)として,インスリン経路遺伝子に注目した,詳細な遺伝子発現の栄養応答解析やRNAiによるノックダウン解析を行い,「特定のインスリン様ペプチドが武器サイズを制御する」ことについて論文発表とプレス発表を行った(Okada et al. 2019, PLOS Biol).また,スクリーニング解析として,比較トランスクリプトームにより,栄養条件や雌雄によって頭部で発現が異なる遺伝子の網羅的探索を進め,多数の候補遺伝子を得ている.さらに,領域の支援および基礎生物学研究所のサポートを受け,10xGenoics社のChromiumシステムを活用して,オオツノコクヌストモドキ(Gnatocerus cornutus)の良質のドラフトゲノム配列を得ることができた.
