Outline of Annual Research Achievements

脊椎動物は陸上への進出に伴い、高濃度の酸素環境に曝されることとなった。本研究では、上陸に伴う酸化ストレスによって、新奇形質を生み出した形態形成プロセスの揺らぎの解明を目指した。ニワトリやマウスのような羊膜類の胚の四肢の形成過程では、指間領域の細胞が細胞死によって取り除かれる「指間細胞死」がおこることで、指が分離する。一方、アフリカツメガエルのような両生類の幼生の肢芽では、指間細胞死のシステムが確立しておらず、それぞれの指が伸長することによって分離する。これらのことから、指間細胞死は脊椎動物が進化の過程で新しく獲得した形質であると考えられている。本研究では、環境酸素濃度に依存して大きくゆらぎ、指間の細胞に特異的に ROS を大量に産生させることで、指間に細胞死を獲得させた経路を解明するために、ROS の産生と、(1) グルコース代謝経路、および (2) 血管形成の関係を明らかにすることを目的として、研究を行った。
まず、(1) グルコース代謝経路については、ニワトリ胚を用いて、① 指間領域のメタボローム解析、② 肢芽におけるグルコース代謝関連遺伝子の発現解析、及び ③ グルコース代謝経路が指間細胞死に与える影響の検証を遂行した結果、肢芽の発生過程では、細胞増殖期から指間細胞死期を通して、酸化的リン酸化によるグルコース代謝が行われていることが明らかとなった。次に、(2)血管網が ROS の産生と関連する可能性を検証した。その結果、ニワトリ胚であっても、アフリカツメガエル幼生であっても、指間において Bmp が発現し、且つ、指間の血管網がリモデリングを行っている時期に、大気中の酸素濃度が十分高いときに限り、指間細胞死が起こっていることを明らかにした (Cordeiro et all., Dev Cell 2019)。
