Outline of Annual Research Achievements

本研究では、花において旺盛な幹細胞の増殖活性を自ら停止し、生殖器官を分化させる仕組みを明らかにするために、これまでの単一変異の解析では見落とされていた遺伝子機能を多重変異体の活用により分子遺伝学的に解明することを主目的としている。シロイヌナズナにおいて、独立に花幹細胞を抑制すると考えられている3つの経路を構成する主要因子:AGAMOUS (AG)、SUPERMAN (SUP)の2つの転写因子と、CLAVATA3 (CLV3) ペプチドリガンドの遺伝学的な相互作用やSUPの作用機構はすでに論文として報告した(Plant Reproduction, 2018; EMBO J. 2018)。さらにCRCによるクロマチンを介したオーキシン合成酵素の制御機構の論文をNature Communications (2018)にて発表した。そこで、我々は、細胞非自立的に働くSUP, CRCタンパク質によるオーキシンシグナルの制御を介した花幹細胞の増殖抑制機構について、特に、2つの転写因子であるSUPとCRCの遺伝学的な相互作用について、二重突然変異体をもちいたオミクス解析の報告を行った。sup crc二重突然変異体は花幹細胞の異常増殖が花発生初期より亢進しており、オーキシン下流において、細胞壁組成やサイトカイニンシグナルの他、代謝経路、ストレス経路が大きく変動していることを明らかにした (Front. Ecol. Evol.2019)。これらSUPとCRCで共通して制御される遺伝子によって、花幹細胞の増殖抑制および正常な数の雄しべ形成がもたらされていることがわかった。
さらに、花幹細胞の増殖抑制時のレビューをInter J. of Mol Sci, 2019やPlant Sig. & Behav. 2019に発表した。
