Outline of Annual Research Achievements

アブラナ科のRorippa aquaticaは、ちぎれた葉の根元側からの再生により繁殖する。この過程には特別な条件は必要なく、水分状況さえ適切であれば、葉の切断に応答して植物体が再生する。これは、葉の一部の細胞が幹細胞性を維持しており、切断などの外的刺激に応答して幹細胞の増殖が再開するからであると考えられる。本研究では、R. aquaticaの葉の断片からの再生をモデルとして、植物の再生や多能性幹細胞の維持・増殖のメカニズムを明らかにすることを目指した。主な成果は以下の通りである。
葉片からの再生と幹細胞性維持機構の解析:再生過程の経時的なRNA-seq解析の結果と、植物ホルモンの網羅的定量解析から、再生の初期にはオーキシンとジベレリン、後期のシュートの再生にはサイトカイニンが重要であることがわかった。また、再生の誘導に必要な遺伝子ネットワークを同定するため、葉からの再生をしないシロイヌナズナとの比較トランスクリプトーム解析を行った。
新奇器官である再生繁殖芽の解析: R. aquaticaの茎生葉の葉柄の基部には、腋芽とは別に栄養繁殖に利用される芽(再生芽)がついており、茎生葉が花茎から離脱すると成長して栄養繁殖する。このような再生芽の存在は珍しいため、形態学的な解析を行った。また、RNA-seq解析により、離脱による器官成長の誘導機構について解析した。
植物の再生促進因子の探索と応用:葉片をMS培地に置床すると、普段は再生してこない葉片の先端部側からの再生が促進されることがわかっていたので、MS培地中に含まれる再生促進因子を同定した。また、ケミカルライブラリーから、植物の再生や幹細胞製の維持に影響を与える化合物を探索するためのスクリーニングを準備した。
なお、当初、学会等への参加のための旅費の執行を予定していたが、新型コロナウイルスの影響で中止になったため、未使用金が生じた。
