Outline of Annual Research Achievements

膜タンパク質および分泌タンパク質は、ERからcis-Golgiに入り、trans-Golgiから出芽した膜輸送小胞に搭載され細胞の周辺部に順行輸送される。神経細胞等で長距離輸送を担う順行輸送分子モーターとしてキネシン-1が知られているが、輸送開始前の膜小胞形成でキネシン-1が果たす役割、キネシン-1が接続する分子および、出芽の際に搭載される分子の選択機構はよくわかっていなかった。研究代表者は、これまでに、キネシンー1受容体としてアミロイド前駆体タンパク質(APP)とアルカデインα(Alcα)を同定し、2019年度までに内容物のプロテオミクス解析や輸送速度制御を行った結果2種は独立性の高い輸送小胞である可能性を示してきた。2019年度に解析を進めた結果、Alcαに優先的に検出された受容体X(未発表のため物質名は伏せる)は、Alcα-KOマウスでは、その一部がAPP小胞によって代替輸送されるが、別の膜タンパク質Y(受容体)は代替輸送を受けない可能性が示された。これら積荷の搭載具合の違いは、ゴルジ体おける選別的出芽ゾーン形成機構を解析するモデルなると考えられた。さらに受容体Xを欠失する細胞で観察される表現型が、これらのキネシン-1受容体欠失の神経で観察できるか解析を行い、ゴルジ体における選択的積荷の選択機構の不調が機能不全に関わることを世界で初めて示しつつある。研究はまだまとまっていないが、輸送小胞形成ゾーンの選択機構の解明を行い、その機能不全が神経機能変化に及ぼす影響の解析を進める。
