Outline of Annual Research Achievements

飢餓時に葉緑体を分解する部分分解オートファジーと、光障害時に壊れた葉緑体を丸ごと除去する全分解オートファジー(クロロファジー)の駆動を司るオルガネラゾーンに着目した本研究計画において、H31-R1年度は以下3項目について成果が得られた。
【1】葉緑体のオートファジー応答ゾーン形成因子の同定:昨年度に同定した光ストレス時の全分解応答ゾーンの候補因子、について、その変異株を用いた解析系を確立し、クロロファジー活性が大きく低下することを確認した。よって当該タンパク質が全分解オートファジーの遂行に強く関わることが明らかとなった。また、飢餓時の部分分解応答ゾーン形成因子を生化学的に同定する解析を行い、膜の形態変化を担う可能性が高いタンパク質が同定された。
【2】葉緑体のオートファジー応答ゾーン形成のイメージングと機能解析:全分解クロロファジー過程のイメージング解析のための多重蛍光可視化系統を整備した。そのタイムラプス観察を行い、全分解ゾーン形成前後のモニタリングを行った。
【4】植物細胞死における葉緑体-核連携ゾーンの機能解析
昨年度に確立した葉緑体-核連携ゾーンのイメージング法を活用し、葉緑体のチューブ状構造「ストロミュール」と細胞死の頻度の変動を調査した。葉の老化が進むと共にこの連携ゾーン形成が活性化され、その後細胞死が起こること、植物ホルモン・サリチル酸シグナルを抑制すると、ストロミュール形成が抑制されることを見出した。よって植物ホルモンと葉の老化進行に応答し葉緑体-核連携ゾーンが形成されることが示唆された。
