小胞体―ミトコンドリア連携ゾーンにおけるII型膜タンパク質CKAP4の機能解析

Outline of Annual Research Achievements

CKAP4は主として小胞体に局在し、N端側を細胞質側に、C端側を小胞体内腔側に向けるII型膜タンパク質であり、小胞体の構造維持に関与することが報告されていた。私共は、2018年度に得た実績に加えてさらに下記の成果を得た。
CRISPR/Cas システムを用いて作製したCKAP4ノックアウト(KO)細胞において、ミトコンドリアのクリスタの形態異常、膜電位の低下、呼吸能の低下、小胞体-ミトコンドリア接触点の増加、ミトコンドリア内のカルシウム濃度の上昇、低グルコース条件下での細胞増殖の低下を認めた。CKAP4の新規結合タンパク質として同定したVDAC2(ミトコンドリア外膜に存在するチャネルタンパク質)は、パルミチン酸化を受けない変異体CKAP4C100Sと結合しなかった。VDAC2は小胞体-ミトコンドリア接触点において、イノシトール三リン酸受容体(IP3R)やGRP75と相互作用し、小胞体からミトコンドリアへのCa2+の流入に関与する。CKAP4 KO細胞で観察されたミトコンドリアの機能異常や小胞体-ミトコンドリア接触点の過形成は野生型CKAP4ではレスキューできたが、VDAC2と結合できないCKAP4C100Sではレスキューできなかった。一方、CKAP4C100Sは野生型CKAP4と同様に、小胞体-ミトコンドリア接触点と非接触点の両分画に局在した。さらに、CKAP4 KOマウスは長期飼育を続けると生後一年半頃から軽度の運動失調を示し、小脳プルキンエ細胞の部分的脱落を認めた。
これらの結果から、CKAP4のパルミチン酸化はCKAP4の小胞体における局在制御よりも機能制御に重要であり、CKAP4はVDAC2との結合を介して、ミトコンドリアの機能を調製することが示唆された。
