糖鎖修飾によるインテグリンの選別輸送ゾーンの制御とその分子機序の解明

Outline of Annual Research Achievements

N-型糖鎖に主に働く三つのα2,3シアル酸転移酵素(ST3Gal3、ST3Gal4とST3Gal6)は、それぞれ異なる膜受容体を特異的に修飾することを明らかにした。我々は、CRISPER/Cas9システムを用いて、それぞれ酵素欠損(KO)細胞株を作成し、インテグリンβ1やEGFRをモデル標的分子として詳細に検討した。興味深いことに、β1インテグリンのα2,3シアリル化はST3GAL4-KO細胞で低下するのに対して、EGFRのα2,3シアリル化はST3GAL6-KO細胞で低下した 。さらに、ST3GAL6-KO細胞に3つの遺伝子をそれぞれ過剰発現したところ、ST3GAL6の過剰発現のみがEGFRのα2,3シアリル化と細胞形態を回復させることができた。同様に、ST3GAL4-KO細胞にST3GAL4を過剰発現させた時のみ β1のシアリル化が回復した。即ち、異なるα2,3シアル酸転移酵素が同じ細胞内に存在していても、相補していないことを示している。これらの結果は、3種のα2,3シアル酸転移酵素がそれぞれ固有の役割をもち、異なる標的タンパク質を修飾していることを明確に示している。また、インテグリンα3とα5鎖のα2,3シアリル化は異なるシアル酸転移酵素によって修飾されることも分かった。今後、トランスゴルジネットワーク(TGN)におけるシアル酸転移酵素と膜受容体の共局在や動態とその機序の解析が必要である。
また、我々は、コアフコースが膜受容体FLT3の二量体形成や細胞内シグナルを強く制御することも見出した。現在、 FLT3の細胞表面への輸送機構を含めた分子機序を解析している。
