Outline of Annual Research Achievements

最近の脳機能イメージング研究は、大脳基底核に加えて、小脳の活動異常がパーキンソン病(PD)に関与する可能性を示唆している。本研究では、PDの霊長類モデルを用いて、大脳皮質、大脳基底核、視床、および小脳を含む広範な神経ネットワークから単一ユニット活動および局所電場電位の同時記録をおこなう多領域多点同時記録を実施し、これらの領域の相互作用を特に周波数成分に着目して解析することにより、PDの病態発現への小脳の関与を明らかにするとともに、小脳への脳深部刺激(DBS)によるPDの治療効果を検討することを目的とした。
研究代表者らは、これまでMPTP投与によって作製したPDサルモデルから、安静時およびボタン押し課題遂行中における一次運動野、大脳基底核(主に淡蒼球)、小脳(主に小脳皮質)から神経活動の多領域多点同時記録を実施した。その結果、PDサルモデルの小脳からベータ波の過活動を検出し、更にcross-frequency coupling(CFC)解析により、運動遂行時における一次運動野との間のphase amplitude coupling(PAC)が大脳基底核よりもむしろ小脳で顕著であることが明らかにした。具体的な結果は次のとおりである。
(1)時系列に基づいて、淡蒼球と一次運動野との間のCFCを解析したところ、健常時やチックモデルではベータ帯域におけるPAC が運動遂行時に強く検出されるのに対して、PDモデルでは同様のCFC現象がほとんど消失していた。
(2)同様に、小脳皮質と一次運動野との間のCFCを解析したところ、上記(1)の結果と異なり、健常時やチックモデルにおいて運動遂行時にみとめられるベータ帯域でのPAC が、PDモデルにおいても検出された。
以上の結果は、PDの病態発現における小脳の関与を示唆しており、現在、サル2頭分のデータをまとめて原著論文の発表を検討している。
