Outline of Annual Research Achievements

地球人口の約95%に、ヒトヘルペスウイルス(HHV)は潜伏感染している。地球上では、HHVと人類の可塑性(免疫)は平衡関係にあり、健常人がHHVにより深刻な病態を引き起こすことは稀である。しかしながら、宇宙は潜伏感染ウイルスと人類の絶妙な平衡関係が破綻するリスクに富む空間であると予想される。また、宇宙飛行士のHHV再活性化という事例を契機とし、アメリカ航空宇宙局は、宇宙飛行時、免疫系の変化を早期に察知するために、HHV再活性化のモニタリングが有効であると報告している[NASA Tech. Innov. 15 (2010)]。したがって、宇宙開発上、HHV再活性化状態の定量的かつ高感度な検査キットの確立や、HHVが潜伏感染を成立・維持させる機構、極限ストレス(重力、睡眠障害等)下における潜伏感染症の推移に関する知見をあらかじめ蓄積することは重要な課題であると考えられる。しかしながら、一連の研究課題の大部分は未着手なままであった。そこで、本研究では、代表的HHV、単純ヘルペスウイルス(HSV)に注目し、高感度有機小分子蛍光プローブを用いた定量的なHSV検査試薬の開発、HSVの潜伏感染成立・維持機構の解明、さらには、極限ストレスや加齢ストレス下におけるHSV潜伏感染の推移に関する知見の解明を試みた。
本年度は、宇宙ストレスとHHV再活性化に関する知見を要約した総説(NPJ Microgravity. [in press])を報告した。また、HSVの糖タンパク質gBを標的する高感度有機小分子蛍光プローブを用いた検査試薬の合成を試み、実験室レベルではHSV感染を検出可能な低分子の創生に至った。加えて、簡便かつ敏速なHSV再活性化マウスモデルの構築にも至った。本実験系は、今後、HSVの潜伏感染機構の解明を試みる上で、強力な研究ツールとなることが期待される。
