臓器間のDNA損傷耐用能の違いから迫る「宇宙放射線克服ストラテジーの基盤構築」

Outline of Annual Research Achievements

平成30年度に作製したマウスモデル、すなわちゲノムDNAの二重鎖切断部位をGFP fociとして可視化できる遺伝子改変マウスに対して、宇宙放射線を模した各種放射線を量研機構・放医研で照射し、in vivo光イメージング実験と免疫染色実験を実施、継時的にDNA損傷の頻度を定量した。また、同遺伝子改変マウスの各臓器におけるDNA損傷修復能と放射線種との関係を解析した。同時に、遺伝子改変マウスから調整した培養細胞を用いてin vitroの実験も実施した。その結果、ガンマ線と鉄イオン線に対する細胞のレスポンスが臓器ごとに異なるさまが見て取れた。具体的には、ガンマ線と鉄イオン線によって生じるDNA損傷の程度と損傷が生じるタイムコース、およびDNA損傷修復のタイムコースが、臓器によって異なっていた。
独自に同定した放射線感受性・抵抗性誘導能を左右する新規遺伝子(HISP2)に着目して、その作用機序を解明する分子細胞生物学実験を展開した。その結果、HISP2が細胞から分泌され、オートクリン・パラクリン的に作用して、HISP2を受容した細胞のEGFRシグナル伝達経路を活性化すること、またNrf2依存的な抗酸化活性を活性化し、結果的に細胞の放射線抵抗性を亢進することを明らかにした。また、HISP2を過剰発現した場合に、確かに細胞の放射線抵抗性が亢進することを明らかにした。さらに血中を循環するHISP2タンパク質を定量する実験系を確立し、同タンパク質の濃度を指標に放射線感受性・抵抗性を予測する手法への道を拓いた。
