Outline of Annual Research Achievements

ゼブラフィッシュを宇宙環境で飼育し、脳において発現が変動する遺伝子を同定した。またこれと並行して、ヒトの生理機能とも深く関係する食欲、情動、ストレス、睡眠等を司るゼブラフィッシュ脳の機能的神経回路の機能を、遺伝学、神経活動イメージング等を駆使して解析した。
1.「空腹と満腹が食欲を調節するメカニズム」:我々は、 ゼブラフィッシュ稚魚の摂餌行動中に、視覚刺激が食欲中枢である視床下部下葉を活性化する神経回路を明らかにしてきた(Muto et al. Nature Communications 2017)。今回、視床下部下葉の活性が満腹、空腹に応じて調節されることを神経活動イメージングにより見出した(Wee et al. eLife 2019)。この成果は、過食症や摂食障害などのメカニズムの理解と治療の基盤になる。
2.「睡眠時のゼブラフィッシュ脳神経細胞の活動を明らかにした」:ゼブラフィッシュ稚魚の脳神経細胞の活動のカルシウムイメージングを行い、睡眠時の脳の活動が哺乳動物と共通性があることを明らかにした(Leung et al. Nature 2019)。睡眠をコントロールする薬剤をゼブラフィッシュを用いてスクリーニングする可能性を拓いた。
3.「宇宙環境で脳において発現が変化する遺伝子の解析」:計画研究瀬原班と共同で、微小重力環境において発現上昇している遺伝子としてTRPN1遺伝子を同定した。TRPN1遺伝子は先行研究で、重力感知に関与していることが知られている。比較ゲノム解析により、メダカを含む魚類の一群(新真骨類)でTRPN1遺伝子が欠失していることを見出した。メダカは、背光反応を示すが、ゼブラフィッシュは示さない。背光反応は重力と光刺激のバランスで引き起こされると考えられている。ゼブラフィッシュノックアウト変異を作製し背光反応が増強するかを解析中である。
