Outline of Annual Research Achievements

大脳皮質は脳情報処理の中枢であり、ヒトに至る進化の過程で著しく発達してきた。従って、脳情報処理の基盤となる大脳皮質の神経回路の進化のプロセス、高等哺乳動物における大脳皮質神経回路の解析、およびその形成過程の解明は重要である。我々は、発達した大脳皮質を持つ食肉類哺乳動物フェレットを用いて、この問題にアプローチしてきた。ヒトやサルなどの形成過程の大脳皮質には、外側線維層(outer fiber layer)および内側線維層(inner fiber layer)という2つの線維層が存在することが知られていたが、マウスにはこの区分は見いだされておらず、投射先およびその形成過程については不明な点が多かった。我々はこれまでに、子宮内エレクトロポレーション法をフェレットに応用することに成功し、フェレットの大脳皮質での遺伝子発現を可能としてきた。そこで本研究では、フェレット大脳皮質神経細胞へGFPを導入し神経回路を可視化することにより、フェレット大脳皮質の線維層の解析を行った。その結果、ヒトやサルと同様にフェレットにも外側線維層と内側線維層が存在していることを見いだした。投射先を解析した結果、外側線維層は近隣の大脳皮質へ、内側線維層は対側大脳皮質もしくは皮質下へ投射していることが分かった。さらにマウスとフェレットとの比較解析により、これまで報告されていなかったマウスにも外側線維層および内側線維層に対応する線維層が存在すること、また進化の過程で外側線維層の量が著しく増加していることを見いだした。これらの研究の成果は、大脳皮質における脳情報動態や大脳皮質の進化プロセスの解明に繋がる。
