Outline of Annual Research Achievements

本課題では、最新の電子顕微鏡観察技術と画像解析技術を組み合わせて、シナプス結合の微細構造のダイナミズムを精緻且つ定量的に評価する解析系を確立し、「シナプス結合の構築則」を明らかにする。そしてこの構築則に基づいて「シナプス関連分子のシナプス構築における役割」を同定すると共に、「精神疾患モデルマウスにおける新たなシナプス構造異常の検出」も目指す。R1年度に実施した実験と結果の概要は以下の通り。
(1)シナプス内微細構造間相関解析法の強化 R1年度までにFIB-SEMを用いたシナプス前後構造丸ごとの定量的形態解析手法の基盤を確立した。また、「画像解析プログラムの開発」については、AI技術を用いた画像情報の自動抽出に必要な電子顕微鏡画像の必要条件を決定した。そこでR1年度は本解析基盤を応用した下記解析を実施した。
(2)シナプス内微細構造相関解析法の応用 Neuroligin-3 KOマウス、N-cadherin KOマウスのシナプス構造を解析し、Neuroligin-3 KOマウスにはシナプス前後協調の異常が認められないことを明らかにした。また、豊富環境下で飼育したマウスのシナプス構造についても解析し、神経活動とシナプス可塑性の誘導は、シナプス前後協調に変化を与えないことを明らかにした。これらの結果をまとめ、現在論文投稿の準備を行っている。
(3)脳組織の急速凍結法と電子顕微鏡試料作成法の確立 高圧凍結法を利用した物理固定 (氷包埋) 法による試料作製手法を用いて、シナプス結合の微細構造情報を取得する実験を行い、生きた実験動物から迅速に脳組織を採取し、急速凍結する実験プロトコールのテストを数回実施すると共に、凍結した試料を電子顕微鏡観察用に樹脂包埋する方法を導入し確立した。
