Outline of Annual Research Achievements |
グリア細胞が脳機能に果たす役割の重要性が明らかにされつつある。本研究では、アストロサイト(A)、ミクログリア(M)、神経細胞(N)が構築するネットワークをMANネットワークと規定し、これらを正確に「見る」「制御する」ことで、脳情報動態におけるグリア細胞の役割を明らかにした。 1 アストロサイト。アストロサイトはシナプスを取り囲み、自身も周辺シナプスとして機能する所謂「三者間シナプス」仮説は、in situで実際に起きているのかどうかについては不明な点が多い。そこで、先ず神経細胞とアストロサイト機能を、高時空間分解能、同時、in situ、で「見る」Ca2+イメージングシステムを構築した。アストロサイトは膜移行型Ca2+蛍光指示薬Lck-GCaMP3又はGCaMP6で、神経細胞はRCaMP2(尾藤らと共同研究)により、同一標本で解析できる系を構築した。神経活動の一定潜時後に、アストロサイトのCa2+応答を認め、これはATPによる神経→アストロサイトのシグナルであった。アストロサイトにP2Y1受容体を強制発現させてこの応答を「制御」する系を構築し、その様式・機序を明らかにした。これにより、アストロサイト→神経細胞のフィードバックシグナルがグルタミン酸によって伝わっていることも明らかにした。このように、神経-アストロサイト間で双方向性のコミュニケイションが存在していること、そのシグナル分子、シグナル伝達様式をin situで明らかにすることが出来た。 2 ミクログリア。ミクログリアはシナプス再編により神経機能を制御することが良く知られている。先ず、ミクログリアを薬colony stimulating factor1 receptro (CSF1R)により可逆的に除去する実験系を構築した。これを用いてミクログリアがシナプス伝達に与える影響の機能解析を開始した。
