Outline of Annual Research Achievements

線虫C. elegans の2つの行動様式、「ローカルサーチ」と「グローバルサーチ」の違いを特徴付ける行動要素として、後退や急激な方向転換などのターン運動の頻度が挙げられる。すなわち、ローカルサーチモードではターン運動の頻度が上昇し、方向転換を繰り返しながら局所的な探索運動を繰り返すのに対し、グローバルサーチモードでは、方向転換の頻度を著しく減少させることで、より広範な範囲を探索する。最終年度においては、これらの後退などの方向転換運動を制御する神経回路を同定することに成功した。具体的には、行動中の多数の線虫個体をトラッキングするMulti worm tracking システムを駆使して、細胞死を誘導するキャスパーぜやminiSOGなどの遺伝学的手法によって、神経回路への摂動をあたえた集団から行動データを取得した。これらの行動データに対してシミュレーションを用いた解析を行い、ここの行動要素に重要な神経細胞を絞り込むことで、それぞれの行動要素の制御に関与する神経回路を明らかにすることに成功した。これらの解析から、後退や急激な方向転換などの単一の行動要素であっても、線虫が置かれた温度環境に応じて、異なる神経回路が行動制御に関与していることを見出した。また、温度環境に応じた行動制御には、感覚ニューロンと介在ニューロン間の情報伝達様式に変容が生じることが重要であることを明らかにした。これらの研究成果を、とりまとめ、論文発表を行った。(Ikeda et al, PNAS, 2020)。
