Outline of Annual Research Achievements

本研究は、海馬の出力層にあたる海馬台からどのような情報がどの脳領域へ伝達されるかを明らかにすることを目的とした。まずは大規模計測と光遺伝学を組み合わせて、投射先を同定した海馬台の神経細胞の活動を計測する実験系を構築した。具体的には、アデノ随伴ウイルスベクターを用いてラットの海馬台の神経細胞にチャネルロドプシン(ChR2)を発現させた。次に、最大256点の多点電極シリコンプローブを用いて、50個程度の海馬台の神経細胞の活動を一斉に計測しながら、海馬台の主要な投射先に挿入した光ファイバーを使って脳領域特異的に青色光で刺激した。ChR2を発現している神経細胞の軸索で生じたスパイクが短い潜時と小さなジターで逆行性に細胞体へ伝わることを指標にして、海馬台の神経細胞の投射先を同定できた。この技術を使って、行動課題(空間探索課題や作業記憶課題など)時と睡眠中に50個程度の神経細胞の発火活動を一斉に計測し、神経細胞の投射先を同定した。計測した活動データから、オフラインでスパイクソーティングにより個々の神経細胞の発火タイミングを抽出し、データ解析を行った。神経細胞が持つ各種の情報(場所情報・頭方向情報・作業記憶情報など)を定量し、海馬台で観察される各種のオシレーション(シータ波・ガンマ波・リップル波など)に対する発火の相関、各種の行動状態(課題遂行時・安静時・レム睡眠時・ノンレム睡眠時)における発火パターンを精査した。その結果、海馬台の神経細胞は、投射先の脳領域によって異なる種類の情報を伝達し、異なる発火パターンで活動し、各種のオシレーションに対して異なる相関を示すことが分かった。本研究により、海馬台が投射先選択的に特有の情報を特有の活動様式を用いて伝達することが明らかとなった。
