Outline of Research at the Start

光合成生物の集光性色素タンパク質は、光エネルギーの捕集と過剰エネルギーの散逸という二つの役割を担う。中でも、チラコイド膜内外で生じるプロトン濃度勾配に依存した光エネルギー消光は、緑藻や植物といった緑色進化系統では広く研究されているが、珪藻や渦鞭毛藻といった赤色進化系統においては研究例がほとんどない。珪藻の集光性色素タンパク質FCPは、タンパク質組成や色素組成が植物の集光性色素タンパク質LHCと大きく異なるため、光捕集・散逸機構が異なると考えられている。本研究は珪藻におけるプロトン濃度勾配に依存した光エネルギー伝達メカニズムを明らかにする。

Outline of Annual Research Achievements

集光性色素タンパク質(LHC)は、光エネルギーを捕集し、光化学系タンパク質(PSI、PSII)へと伝達する重要な役割を担う。LHCは光合成生物種間で多様であり、タンパク質構造の違いに加え、色素分子の種類や数の違いも見出されている。LHCの多様性は、光合成生物の見た目の色の違いをもたらし、励起エネルギー伝達機構の違いを担う。本研究では褐色を呈する珪藻を材料とし、珪藻の特殊なLHCであるフコキサンチンクロロフィルタンパク質(FCP)の機能構造解明を目指す。
クライオ電子顕微鏡単粒子解析により、珪藻のPSI-FCPI超複合体の立体構造を解明した。その結果、PSIには遺伝子の異なる16個のFCPが結合していることを見出した。陸上植物のPSIには4個のLHCが結合しているため、珪藻と陸上植物とでPSIのアンテナサイズが大きく異なることを明らかにした。この結果を論文として纏めた。
培養条件の変化がFCPの発現に及ぼす影響を調べた結果、温度および二酸化炭素濃度を変化させることにより、FCPの発現調節が変化することを見出した。特に、30度、3%CO2の培養条件において、新規のFCPが発現した。この結果を論文として纏めた。
強光照射した珪藻からFCPを調製し、励起エネルギーダイナミクスを調べた。強光により、状態の異なるFCPが発現し、励起エネルギー消光を誘導することが判明した。光合成生物は光の強さを感じると光阻害を起こす。光阻害が進行することによりチラコイド膜のルーメン側が酸性化される。強光およびpH変化によってFCPの状態が変わり、励起エネルギーダイナミクスの変化が生じたのであろう。この結果を論文として纏めた。
