Outline of Research at the Start

本研究では、光合成調節因子であるCP12分子群の分子生物学的役割を分子集合レベル、タンパク質構造レベル、および速度論的に解明し、C4光合成調節の分子機構モデルを構築する。申請者は、これまで光合成カルビン回路の調節因子であるCP12の構造生物学的な先駆的研究を行ってきたが、最近、共同研究者らによって、CP12様タンパク質CP12-3がフラベリアにおけるC4光合成の重要因子であることが分かった。本研究では、CP12分子群が形成する複合体の解析を進め、環境変動に応答するための光合成調節機構の分子機構モデルを構築する。

Outline of Annual Research Achievements

本研究では、C4光合成の律速因子として同定されたCP12-3を研究対象として、(1)CP12-3の構造機能解析を実施した。それに加えて新学術領域の他の研究者と共同で(2)光合成関連酵素群の構造機能解析を行った。以下に詳細を述べる。
(1)については、フラベリアCP12-3の発現・精製、ならびにCP12-3の結合標的候補の酵素としてMDH, NADP-ME, PRK, GAPDHの発現・精製を試みた。CP12-3, MDH, NADP-MEについては大腸菌での発現・精製系構築することができた。さらに、これまで達成できてこなかったGAPDHについては、CP12-3と共発現することによって、大腸菌での発現・精製系を構築することができた。その後、CP12-3とGAPDHが相互作用することがわかったため、CP12-3とGAPDHの相互作用解析を実施した。その結果、CP12-3がGAPDHの非特異的な集合を抑制する可能性を見出だすことができた。さらに、CP12-3/GAPDH複合体、およびGAPDH単体の構造解析に取り組んだ結果、GAPDH単体の構造解析に成功した。通常、四量体で存在するGAPDHが二量体で存在することが明らかとなり、この結果からCP12-3の新しい機能、すなわちGAPDHの構造安定化に寄与する可能性が示された。
(2)については、ソルガムRubiscoのRbcSをイネに導入し、さらにCRISPR-Cas9で野生型イネ(Nipponbare)のRbcSをノックアウトしてハイブリッドRubiscoを作成したところ、酵素活性が1.9倍、光合成活性も1.2倍程度上昇することが分かった。その成果を最近論文として発表することができた(Mol Plant, 13, 1570, 2020)。現在は、この分子機構を元に、ソルガムとは異なるRbcS遺伝子を導入した改良型イネの研究を展開し、さらに詳細な分子機構がわかりつつある。
