Outline of Research at the Start

「システムウイルス学（実験ウイルス学と他分野の学際）研究」という本研究の独創的なフレームワークは、既存のウイルス学に留まらず、「（宿主and/orウイルスの）進化」という地球史における生命現象の謎の根幹にアプローチできる研究体系であり、領域内で協同することで相互発展的な進展を期待できる点が、本申請最大の特色である。

Outline of Annual Research Achievements

計画1 ヒト集団におけるウイルスの適応進化原理の分子メカニズムの解明
HIV-1B, HIV-1Cそれぞれの臨床分離株を基に、PTAP欠損、YPxL欠損、両欠損変異体を作製した。ウイルスの出芽効率および感染価を、培養細胞を用いた実験で検証した。 また、公共データベースLANL HIV Immunology Databaseを用い、PTAP/YPxLモチーフを含むp6ドメインをエピトープとして提示できるヒト白血球抗原(HLA)のハプロタイプ「p6 HLA群」を抽出した。次に、世界中で臨床分離されたHIV-1配列をまとめた公共データベー スLANL HIV Sequence Database、および、世界中の人種のHLA分布をまとめた公共データベースHLA Databaseをそれぞれ用い、世界各国におけるPTAPを重複した株、YPxLを欠損した株の割合、および、世界各国におけるp6 HLA群の頻度、をそれぞれ抽出した。その後、生物統計学的解析により、これらの頻度に相関があるかどうかを検証した。以上より、「アフリカ特異的な獲得免疫の選択圧によって、PTAPモチーフの重複、および、YPxLモチーフの欠失が誘導された可能性」について検証した。現在論文化を進めている。

計画2 内在性レトロウイルスとほ乳類の共進化原理の分子メカニズムの解明
"DIGSツール"を用い、ゲノムが解読されている164種のほ乳類すべてにおける、APOBEC3遺伝子と内在性レトロウイルス(ERV)を網羅的に同定した。これにより、ほ乳類の科・属・種において、特異的あるいは保存された遺伝子およびERVを特定した。さらに、ゲノム配列が詳細に解析されている種については、これらのコード領域をすべてゲノム上にマッピングした。以上の研究成果を論文化した（Ito et al, PNAS, 2020）。
