Outline of Research at the Start

研究代表者は、特殊な核酸構造体である「グアニン四重鎖」を形成し、シナプス機能維持のために必要なタンパク質をコードするmRNA群、つまり「シナプスmRNAグアニン四重鎖」を多数同定し、グアニン四重鎖の形状変化によってそれらのシナプスへの輸送や翻訳効率が制御されることを見出した。しかしながら、mRNAグアニン四重鎖構造異常による脳機能への影響については未だ明らかにされていない。そこで本研究では、「精神疾患の認知障害」とシナプスmRNA輸送・翻訳において重要な役割を担う「シナプスmRNAグアニン四重鎖」の因果関係を個体レベルで解明することを目的とする。

Outline of Annual Research Achievements

本研究は、「精神疾患の認知障害」とシナプスmRNA輸送・翻訳において重要な役割を担う「RNAグアニン四重鎖」の因果関係を解明することを目的とした。研究代表者は、未だ生物学的機能が不明である「グアニン四重鎖」に着目し研究を行っている。グアニン四重鎖はグアニンが豊富な配列領域でDNA・RNAが形成する特殊な核酸高次構造のひとつである。研究代表者は、精神疾患モデルマウスを用いた解析で、グアニン四重鎖の構造異常が精神障害を引き起こすことを明らかにした (Shioda et al., Nat. Med. 2018) 。しかしながら、mRNAグアニン四重鎖と「精神疾患の認知障害」との関連性は未だ明らかにされていない。本研究では、グアニン四重鎖を形成し、神経機能維持のために必要なタンパク質をコードするmRNA群、つまり「神経細胞におけるグアニン四重鎖形成mRNA」を多数同定することができた。さらに、それらの中で特に神経機能に重要な役割を担うmRNAを特定し、そのmRNAで形成されるグアニン四重鎖の高次構造を明らかにした。そして、グアニン四重鎖の形状変化によってそのRNA動態が制御されることを見出した。さらに、神経細胞に高発現する新たなグアニン四重鎖結合タンパク質を同定した。そのタンパク質はストレス顆粒の形成に重要であり、神経細胞死に関与することが本研究により明らかになった。この新規グアニン四重鎖結合タンパク質はRNA代謝に影響を与え神経細胞死を誘導する可能性が示唆された。
