Outline of Research at the Start

統合失調症や薬物依存症などの精神疾患は、通常見られない行動により定義・診断されるが、何故異常な行動が生じるのかは分かっていない。そこで本研究では、まず精神疾患の行動表現型を説明するマクロスコピックな神経回路動態を、情報学的手法で同定・モデル化する。次に同定したマクロ回路動態を直接操作することで、マクロ回路動態と精神疾患の行動表現型との因果関係を明らかにする。さらに遺伝子やシナプスレベルの変化が、当該マクロ神経回路動態に与える影響を明らかにする。こうして精神疾患の行動表現型の発現機構を構成的に理解する。このストラテジーは主に動物実験およびin silicoシミュレーションで実施する。

Outline of Annual Research Achievements

統合失調症や薬物依存症などの精神疾患は、通常見られない特有の行動により定義・診断される。精神疾患には遺伝的な要因もあるが、何故遺伝子やそれに伴うシナプス伝達など下層の変化が精神疾患特有の行動表現型に至るのかは分かっていない。これは遺伝子やシナプスの階層と行動層との解離が大きく、論理的な仮説を設定しづらいためであると考えられる。この問題を解決するためには、行動層の直下にあるマクロスコピックな神経回路ダイナミクス（脳活動パターン）に注目して、その行動表現型との因果律を解明すると共に、当該脳活動パターンと下層との関係を解析する戦略が有効である可能性がある。そこで本研究では、まず精神疾患の行動表現型を説明する脳活動パターンを情報学的手法で同定・モデル化し、同定した脳活動を操作することで、脳活動パターンと精神疾患の行動表現型との因果関係を明らかにすることを第一の目的とした。さらに遺伝子・シナプス・メゾ回路層における変容が当該モデルに与える影響を定量化することで、精神疾患の行動表現型が発現するメカニズムの構成的理解を目指す。初年度は、まず精神疾患症状様行動のモデルとしてラットに文脈・感覚入力依存的に意思決定を行わせるオペラントタスクを確立し、当該タスクを実行中の脳活動から行動選択特異的なパターンを機械学習を用いて抽出する技術を確立した。その成果を受け、最終はさらに以下の項目を達成した。

1. 遺伝子・シナプス層変容のモデルとして、統合失調症・薬物依存症モデルラットを導入した。
2. 当該疾患モデルにおいて、上記タスク実行中の多脳領域脳活動（電気生理）記録を行った。
3. 取得した脳活動記録から、疾患症状および行動選択特異的なパターンを抽出・モデル化した。
4. 抽出・モデル化した脳活動パターンに化学遺伝学的に介入することで、当該パターンと疾患表現型との因果関係を証明した。
