Outline of Research at the Start

幹細胞は通常の体細胞より全遺伝子の転写活性状態が極めて高いHypertranscription(高転写状態)にあることが知られている。中でも成熟卵母細胞、精原細胞の胚性前駆細胞である始原生殖細胞の解析は最も先駆的に行われている系の一つである。細胞分化制御機構の解明とその人為的な操作に向けて、従来の転写因子による遺伝子発現制御に加えて、新たに遺伝子発現の“量的”制御の解明が期待されている。本研究では、高転写状態の始原生殖細胞をモデル系として、高転写状態の意義や獲得メカニズムの解明のための新規技術開発を行う。

Outline of Annual Research Achievements

転写産物の絶対量測定の精度が増し、細胞分化において、ゲノムワイドに転写活性化状態が変化する発見や報告が相次いでいる。中でも古くから知られている未分化段階の細胞に認められるHypertranscription、高転写状態は、受精卵のZGA、造血幹細胞分化において特に解析されてきた。これら幹細胞状態の細胞では、ゲノム全体が遺伝子によらず須らく高転写状態になっており、分化の過程で、通常の転写状態に戻ることが明らかになっている。そこで、高転写状態の解明には、転写制御をクロマチン構造のレベルから理解することが不可欠である。すなわち、組織内で、同一の細胞内のクロマチン構造解析とトランスクリプトーム解析を“1細胞レベル”で同時に行う必要がある。そこで本研究では、生殖細胞及びその分化段階での転写絶対量測定とクロマチン構造解析を1細胞レベルで同時に行うことで、高転写状態の定量とクロマチン構造レベルの形成メカニズムの解明を目指した。具体的には、生殖細胞を含む固定化組織切片を用いて、mRNA量の絶対量と転写を制御するクロマチン構造状態(転写因子・クロマチン制御因子の結合、ヒストン修飾・バリアント置換、各活性化状態におけるRNA polymeraseIIのゲノム領域へのリクルート)を同時に測定する技術を開発した。本手法では、単一細胞レベルで転写量総量の測定とエピゲノムの解析を同時に行うことが可能であり論文投稿を進めている。また、高転写状態を解明するためには翻訳産物であるタンパク質の同時測定が欠かせない。そこで、これらエピゲノム、mRNAに加えてタンパク質を同時に測定する技術の開発を進め、合わせて論文投稿準備を進めている。
