Outline of Research at the Start

近年のイメージング研究の進展により、同じ種類の細胞であっても生体の中で異なる振る舞いを示す 「細胞間不均一性」が明らかになってきた。申請者は、細胞の動態データから細胞間不均一性をバイアスなく図示し、細胞間の違いを司る軸を明らかにするデータ解析方法を開発した。 本研究では、この申請者の研究に立脚し、マウスの in vivo イメージングの細胞データに対して、外れ値となる免疫細胞を同定する。その動態を詳細に解析し、細胞分布の経時変化を解明して、外れ値免疫細胞の存在意義を明らかにする。

Outline of Annual Research Achievements

多光子顕微鏡で観察された免疫細胞の運動を解析する過程で得られた、距離学習による細胞軌跡の可視化手法を、Springer社の国際学会特集書籍に原著論文として投稿し、査読を経て掲載された。(Nakamura N and Yamada R. Proceedings in Mathematics & Statistics (Methods of Mathematical Oncology: Fusion of Mathematics and Biology): Distance-matrix-based extraction of motility features from functionally heterogeneous cell populations, 283-293, Springer (2021).)同様の距離学習は、細胞間の不均一性以外のみならず、個体間の表現型の不均一性を可視化する目的で利用可能である(Nakamura N et al., medRxiv 2022)。近年盛んになっている深層学習ベースの学習方法とは異なる、新たな教師なし学習手法として拡張可能性があると考えられ、今後も研究を進めていく。また、複数の数理モデルの距離を木構造を用いて比較する新規方法について、共同研究者と進めていた研究が情報系の国際学会 ISAAC 2022 (33rd International Symposium on Algorithms and Computation) に査読を経て採択され、Leibniz International Proceedings in Informatics に収録された。(Akutsu T, Mori T, Nakamura N et al. On the complexity of tree edit distance with variables. doi: 10.4230/LIPIcs.ISAAC.2022.44)この手法についても、上記の距離学習の副産物というべき成果であり、BioModelsなどの数理モデルデータベースへの応用を検討している。
